Handlingen följer Frederick Forsyths roman med samma namn närmast fullständigt, och skildrar hur lönnmördaren med täcknamnet Schakalen (Edward Fox) av OAS högsta ledning får uppdraget att likvidera Frankrikes president Charles de Gaulle. Inom kort får dock den franska säkerhetstjänsten vetskap om att illdådet planeras och sätter sin bästa detektiv, Claude Lebel (Michael Lonsdale), på uppdraget att spåra upp och omhänderta mördaren. Till sin hjälp får han obegränsade resurser ställda till sitt förfogande.

Following 'Unbreakable' (2000) and 'Split' (2016), security guard-turned vigilante David Dunn (Willis) hunts down Kevin Wendell Crumb (McAvoy), a disturbed and dangerous being with 24 distinct personalities. Both men are captured and placed in an asylum under the care of psychiatrist Dr Ellie Staple (Sarah Paulson), who specialises in treating those who believe they have supernatural gifts. However, their fellow patient Elijah Price aka Mr Glass (Jackson) enlists Kevin's help to escape the facility and realise his ambition of revealing their supernatural gifts to the world.

The film features three stories told by different reporters for the supplement magazine for the Kansas Evening Sun newspaper run by Arthur Howitzer Jr. (Bill Murray). The first story, covered by J.K.L. Berensen (Tilda Swinton), takes place in prison where murderer Moses Rosenthaler (Benicio Del Toro) grows increasingly infatuated with prison guard Simone (Léa Seydoux). To demonstrate his feelings, Moses paints an abstract nude of her which piques the interest of art dealer Julien Cadazio (Adrien Brody). In the second segment, reporter Lucinda Krementz (Frances McDormand) covers the student revolutions taking place on the streets of Ennui, where she profiles self-proclaimed leader Zeffirelli (Timothée Chalamet). In the third segment, food critic Roebuck Wright (Jeffrey Wright) recalls the private dinner he attended with the hierarchy of the Ennui police forces which was overshadowed by the kidnapping of the Commissaire (Mathieu Amalric)'s son.
