När Jake Blues (John Belushi) släppts ur fängelset, beger sig han och hans bror Elwood (Dan Aykroyd) iväg för att besöka barnhemmet där de växte upp och uppfostrades av nunnor. <br/>De får reda på att kyrkan dragit in sitt stöd till barnhemmet, som kommer att säljas om inte fastighetsskatten betalas inom elva dagar. <br/>Bröderna beslutar att de ska skramla ihop pengarna genom att samla det gamla bluesbandet och riva av ett gig. De kanske är på ett "uppdrag från Gud" men de skaffar sig hela tiden nya fiender.

Axel Foley reser tillbaka till Beverly Hills där han får en ordentligt otäck berg- och dalbanetur i nöjesparken Wonderworld! Axel får sällskap av sina gamla polare Billy Rosewood och Serge och han blir nöjesparkens häftigaste nytillskott när han jagar skurkar i åkattraktionerna, på showerna och i den underjordiska labyrinten under parken.
